2014 350 Mag Mercruiser showing fault code 2144-6. I understand it is saying not firing on cylinder 6. I checked the spark plug and gap, looks good. Plug wire secured good. Did not pull distributor and rotor yet. Outside of those, what else could cause this. I have the Mercruiser Vessel View on my boat which I really like.

The 2144 is going to be the fault code... The 6 (in your case) is the fault type code number... Universal Failure Condition... Description...

2144Engine misfire6FaultedThis is for both cylinder banks (all cylinders).
Number 6 = Faulted = The device, hardware, a calculation, or a process has failed a rationality check or has detected a fault.

So in your case it is not cylinder number 6....All the cylinders are affected.
You'll want to start off with a new cap and rotor as that is the most suspected issues with this design distributor...Looks good and actual good are 2 different things for this cap... Also be sure the cap and rotor are seated correctly and the hold down screws / threads are not damaged as the housing is plastic.... Next try new spark plugs... Has any service work been done recently? or is this all of a sudden?.. Do you hear / notice a misfire in the engine?...< Meaning possibly fake...

This code has come up online a few times when searched so the following are of some other thoughts but please know one is for a 4 cylinder, and it was suggested cylinder number 4 is the issue but that shouldn't be the case as the code is for all cylinders according to a service manual like noted above....If you can't fix it with a tune up and the engine internals are good, then you may need to reach out to Mercruiser to see of you need an update in the PCM by a service dealer like mentioned in the sea ray thread below.

Also there's this,

Also there's this,

Okay, all plugs, distributor, and rotor replaced. They definitely had wear. Still getting same fault message but now it shows a “7” in the upper right corner. The fault detail page is the same. Is this giving specific info or still a general engine misfire?

7DirtyThe signal following a switch is below the expected value.

Check all power and grounds also suspect crankshaft and camshaft sensors or possibly the in gear indicator switch just to name a few… that all mentioned sounds like it’s possibly time to get the engine scanned as well… hopefully there’s no internal engine damage to the cylinders or valves, etc. might want to use a scope to check inside each cylinder with the spark plugs pull out.
