Will a coil not spark under compression? The plug fires great. When I replace the plug that cylinder is dead?

Some plugs fail under compress

Some plugs fail under compression. Swap plug with other one and you'll see action. Had same issue with a bike. Cost plenty $$$ for $3 fix.
