I'm replacing my fuel/water separator myself for the first time going into my Chrysler 360's. Any tips? Should I fill the canister with fuel before reinstalling? Do I need to vent the top after starting?

Start your engine first and shut it down.
This primes the carburetor, and will make re-starting on a new cartridge easier.

Yes.... carefully fill the cartridge about 80% or so.
Put an oil film on the gasket surface.

I never fill mine. Warm up engine first then change filter, restart and run at a high idle for several minutes. Take a flashlight and look down in the old filter to see what you've collected, mine are always clean.
