Hoses onto clamped fittings - no

threaded fittings (pipe threads) - yes, PST

Gaskets between two machined surfaces - PMR = per manufacturers recomendations. usually doesn't hurt. If unspecified, permatex #3 is usually a good choice.

If you can refine the question a bit, you can probably get a definitive answer."

It was meant to be a rather open question. In the service manual (454XLi), I have not seen a reference to using a sealant for anything water related. Growing up working on my own car, I always used a sealant (usually shellac).
